The double-antibody sandwich method is a type of sandwich ELISA. The antiserum containing the known antibody is adsorbed in a small well on the microtitre plate; then the antigen to be tested is added, and if the two are specific, binding occurs, and then the enzyme-linked antibody that reacts specifically with the antigen to be tested is added, so that one unit of antigen binds two units of antibody at the same time to form a "sandwich". Finally, the substrate of the enzyme is added, and the content of the antigen to be tested is determined by the color of the resulting-colored enzymatic product.
